Scientists at Duke University in the USA have found that the use of marijuana changes sperm in such a way that future offspring may develop obvious anomalies affecting memory, mood and learning processes.
Experiments on laboratory rats have shown that exposure to marijuana on males prior to mating resulted in abnormalities in offspring similar to those caused by tobacco smoke and pesticides. There are few studies of this kind: most of the work on the effects of marijuana on future offspring focuses on pregnant mothers.
"Our results can be transferred from rats to humans because [both] have the same neural circuits," says lead author Theodore Slotkin. - This study demonstrates that use of marijuana by fathers, not just mothers, can affect the health of offspring by taking it before conception".
Male rats were injected with tetrahydrocannabinol (the main psychoactive substance of marijuana) for 28 days at concentrations of two and four milligrams per kilogram of weight, respectively. This is about half the dose (per kilogram of weight) a person gets when smoking a hemp cigarette. Two days after the last injection, the males were mated with females who were not injected with THC. Brain development in offspring of such rodents was studied from 30 days to 150 days.
Tetrahydrocannabinol led to a disturbance in the synthesis of acetylcholine, an important neurotransmitter. Acetylcholine is responsible for synaptic plasticity - the ability of the synapse to increase or decrease the release of the neurotransmitter in response to an increase or decrease in its activity. This phenomenon underpins learning and memory formation. The influence of tetrahydrocannabinol is due to the dose-dependent deficit of binding hemiholinium-III - a molecule that disrupts the transport of the acetylcholine precursor and thus inhibits its synthesis.
